We do layout mockups and interaction workflows in OmniGraffle and comps in Photoshop. Then we slice up the images, put them on an iPad with Dropbox, and build the interactive mockup right on the device with an app called Blueprint: http://itunes.apple.com/us/app/blueprint/id405203705?mt=8

(Blueprint also has a bunch of standard widgets built in, but we usually end up making custom widgets.)

I have been using Antetype for a while now and am quite happy with it. Automatic/flexible layout system, consistent styles with widgets - it's easy to create lots and lots of individual screens and still keep everything consistent.

Just in case you are looking for an idea for which to make an app for the iPad, could you please consider the following?

Scanning a sequence of QR codes that are each made so as to contain just one Unicode character so that a telephone number can be built up without needing to press any buttons.

I have recently produced the following two documents.

a_simulation_about_an_idea_that_would_use_qr_codes.pdf

experimental_table_top_for_telephoning_1200_by_800.pdf

The pdf documents are available as attachments in the following forum thread.

I'm using App Cooker on the actual iPad itself to build interactive mockups and then App Taster to run it on iPads and iPhones without the parent program.

Works pretty well.

Also:
The following is a large spreadsheet comparing all the UX / IXD tools I could find - Everything from Wireframing to Prototyping, on the Mac, PC, and Web.
